The outline of Escherichia coli K-12 W3110 and MGF-01 proteome
We have analyzed the proteome of Escherichia coli K-12 W3110 and reduced-genome strain MGF-01 grown under the M9-glucose medium in the following conditions.
1. Shotgun proteomics (SDS-PAGE in combination with LC-MS/MS)
- (1) Logarithmic phase
- (2) Early stationary phase
- (3) Late stationary phase
- (4) Death phase
2. Peptide Mass Fingerprinting (two-dimensional PAGE followed by MALDI-TOFMS)
- (1) Logarithmic phase
- (2) Early stationary phase
- (3) Late stationary phase
- (4) Death phase
3. N-terminal amino acid sequencing by the protein sequencer
The result of proteome analysis
The number of identified proteins in strain K-12 W3110 amounted to 1412, 1402, 1164 and 1037 in the logarithmic phase, the early stationary phase, the late stationary phase and death phase respectively. 1617 proteins were identified in total.
The number of identified proteins in strain MGF-01 amounted to 1053, 1204, 1087 and 1015 in the logarithmic phase, the early stationary phase, the late stationary phase and death phase respectively. 1368 proteins were identified in total.
In addition, we have also confirmed techniques for reviewing annotations from proteome analysis results. In recent years, such techniques have been termed proteogenomics.
The results of the proteome analysis are available to be downloaded from the DBRP (Data and Biological Resource Platform).
